Utilisation des lipides cellulaires comme indicateurs de pollution dans les milieux naturels : diagnostic intégré d'écotoxicité basé sur des végétaux terrestres et les diatomées des milieux aquatiques

Abstract : The LIPID program aims at assessing the interest of intracellular lipid profiles to improve the ecotoxicity diagnosis in natural environments impacted by phytosanitary practices. It is based on the field-reality of a pilot watershed located in the vineyard of Blayais (basin Livenne Marcillac -33-), already selected as a pilot-site of the PHYTOCOTE program. Two descriptors based on this principle, previously developed by LBM partner on terrestrial higher plants, reveal the toxic effects of micropollutants (especially heavy metals) in soils polluted from lipid profiles of their leaves, and have already been subjected to standardization. A first objective is then to study the transferability, interest and contribution of such a lipid biomarker on the benthic diatom compartment of rivers. We will try to verify that, compared to different descriptors of toxic effects already known on this compartment (growth, structural, functional, transcriptomic descriptors ...), this new biomarker presents interesting characteristics of sensitivity, precocity and if possible , specificity with respect to the toxic mode of action of the tested substances. Robustness of the observed responses with respect to different confounding factors (diatom species, season and water temperature ...) will also be verified. A second objective is to achieve, from the same target vegetal organisms, a comparative approach of the effects of toxic pollutants in a gradient of more or less impacted terrestrial and aquatic natural environments of the pilot watershed. For that, we will use both an in situ approach (direct exposure in the environments which, for diatoms, will mobilize a caging device) and an ex-situ approach (tests performed under laboratory controlled conditions). At the end of the program, it will be possible to evaluate whether the lipid biomarker newly transferred to the benthic diatoms provides a real added value, in terms of sensitivity and robustness, compared to other descriptors of toxic effects positioned at a specific cellular functioning level, or at different levels of biological integration.
