              <p>In mammals, suppressor of cytokine signalling (CISH, SOCS1 to SOCS7) factors control signalling pathways involved in the regulation of numerous physiological processes including pregnancy. In order to gain new insights into the biological functions of SOCS in the endometrium, a comprehensive analysis of SOCS gene expression was carried out in bovine caruncular (CAR) and intercaruncular (ICAR) tissues collected i) during the oestrous cycle, ii) at the time of maternal recognition of pregnancy and at implantation in inseminated females, iii) following uterine interferon-tau (IFNT) infusion at day 14 post-oestrus, iv) following a period of controlled intravaginal progesterone release and v) following transfer of embryos by somatic-cell nuclear transfer (SCNT). The regulatory effects of IFNT on in vitro cultured epithelial and stromal cells were also examined. Altogether, our data showed that CISH, SOCS4, SOCS5 and SOCS7 mRNA levels were poorly affected during luteolysis and pregnancy. In contrast, SOCS1, SOCS2, SOCS3 and SOCS6 mRNA levels were strongly up-regulated at implantation (day 20 of pregnancy). Experimental in vitro and in vivo models demonstrated that only CISH, SOCS1, SOCS2 and SOCS3 were IFNT-induced genes. Immunohistochemistry showed an intense SOCS3 and SOCS6 staining in the nucleus of luminal and glandular epithelium and of stromal cells of pregnant endometrium. Finally, SOCS3 expression was significantly increased in SCNT pregnancies in keeping with the altered immune function previously reported in this model of compromised implantation. Collectively, our data suggest that spatio-temporal changes in endometrial SOCS gene expression reflect the acquisition of receptivity, maternal recognition of pregnancy and implantation.</p>

            <abstract xml:lang="fr">
              <p>Chez les mammifères, la famille des facteurs Suppressor of Cytokine Signalling (CISH, SOCS1 à SOCS7) contrôle les voies de signalisation impliquées dans la régulation de nombreux processus physiologiques y compris la gestation. Afin d’acquérir de nouvelles connaissances sur les fonctions biologiques des SOCS dans l’endomètre, une analyse exhaustive de l’expression des gènes codant pour CISH et SOCS1 à SOCS7 a été réalisée dans les tissus caronculaires et intercaronculaires bovins prélevés (i) au cours du cycle oestrien (ii) au cours de la période de reconnaissance maternelle de la gestation et de l’implantation chez des femelles inséminées artificiellement (iii) après infusion intra-utérine d’interféron-tau (IFNT) à 14 jours post-oestrus (iv) chez des femelles supplémentées en progestérone par diffusion intravaginale (v) après transfert d’embryons produits par transfert de noyau somatique (TNS). La régulation par l’IFNT a également été examinée in vitro à partir de cultures primaires de cellules épithéliales et stromales. Nos résultats montrent que la lutéolyse et la gestation affectent peu les niveaux d’ARN codant pour CISH, SOCS4, SOCS5 et SOCS7. Par contre, les niveaux d’ARN codant pour SOCS1, SOCS2, SOCS3 et SOCS6 augmentent fortement à l’implantation (jour 20 de gestation). Nos modèles expérimentaux in vivo et in vitro démontrent que seuls les gènes codant pour CISH, SOCS1, SOCS2 et SOCS3 sont induits par l’IFNT. Dans le stroma ainsi que dans l’épithélium glandulaire et luminal, une intense coloration des noyaux cellulaires est détectée par immunocytochimie pour SOCS3 et SOCS6. Enfin, l’expression endométriale du gène codant pour SOCS3 augmente chez les femelles gravides porteuses d’embryons TNS, un résultat qui concorde avec l’altération de la fonction immunitaire rapportée précédemment pour ce modèle d’implantation perturbée. Nos données suggèrent que les modifications spatio-temporelles de l’expression des gènes codant pour les SOCS dans l’endomètre reflètent l’acquisition de la réceptivité endométriale, la reconnaissance maternelle de la gestation et l’implantation.</p>
