Search for salivary biomarkers of the period of gilt receptivity to the boar effect

Our objective is to develop alternatives to hormonal treatments for estrus synchronization in gilts. Before puberty, gilts exhibit a “waiting period”, related to ovarian development and gonadotrophin secretions, during which external stimulation, such as boar exposure, could induce and synchronize the first ovulation. Practical non-invasive tools for identification of this period are lacking. We searched for salivary biomarkers of this period. In a previous study, gas chromatography coupled to tandem mass spectrometry (GC/MS/MS) analysis of saliva allowed the identification of two potential biomarkers: 17beta-estradiol (E2) and dehydroepiandrosterone (DHEA), whose concentrations varied at the beginning of the “waiting period”. The aim of this study was to check the efficacy of the boar effect and to determine if E2 and DHEA could be biomarkers of the period of gilts receptivity to the boar effect. Salivary samples were collected from 30 Large-White gilts twice a week from 140 to 180 days of age. Starting at 160 days, 15 gilts were exposed to a boar twice a day and subjected to estrus detection using the standing response to a backpressure test, while the other 15 gilts were subjected only to the standing response test. The percentage of gilts detected in estrus between 161 and 166 days was 71% when exposed to a boar vs 23% without boar. GC/MS/MS analysis of saliva showed no significant difference in E2 and DHEA concentrations between gilts that responded to the boar effect vs gilts that did not. These results confirm that boar introduction stimulates puberty attainment. The relevance of E2 and DHEA as biomarkers could not be demonstrated. Further studies are in progress to search for other biomarkers.
