Amélioration des biotechnologies de la reproduction dans l'espèce asine: congélation de la semence, insémination artificielle, collecte et maturation in vitro d’ovocytes

Abstract : Today, the seven donkey breeds in France are endangered. In the CRB-Anim project, our studies focus on the conservation of genetic material in this species. Our lab hasworked on the development of a freezing method for donkey semen in order to obtain reasonablepregnancy rates (at present, fertility per cycle <15%). The aims were 1) to test extenders developedin the stallion and 2) to study the effect of decrease in temperature in the cryopreservationprocedure,on semen quality. Thus, we demonstrated that donkey spermatozoa need more protectionthan stallion spermatozoathat can be improved by the addition of sterilized egg yolk plasma or by liposomesofegg yolkphospholipids. Moreover, some female specificitieswerestudied:the physiology and the reactivity of jenny cervixhave been evaluated throughout insemination and a technique of oocyte collection using transvaginale ultrasound guided follicular punctureon standing jennyand oocyte in vitromaturation has been developed.
