Etude expressionnelle de gènes impliqués dans la fusion des myocytes au cours de la régénération musculaire chez la truite

Abstract : The muscle is composed of myofibers, having a remarkable growth and regeneration capacity. This capacity require a particular type of stem cells : the satellites cells. These cells generate muscular precursors (myoblasts) that will proliferate in myocytes and merge, either with existing fibers : hypertrophy phenomenon in which the fiber grow, or with other myocytes : hyperplasia phenomenon in which new fibers are formed. The identification of genes involved in the merge of myocytes (Naa15a and Wisp1a) was carried out. My purpose in this internship was to caracterize the kinetics of expression of these genes and the fibers regeneration evolution in a phenotypic point of view, by measuring these genes expression by qPCR and by observing under a microscope some muscular injured fibers, at different times of completion of the regeneration. Our results show that destroyed fibers degenerate during 16 days, and that after their disappearance, new fibers are formed by hyperplasia. Studied genes express themselves from day 16 and keep expressing more and more. In conclusion, our results show that during a muscle injury in fish, the muscle regenerate by new fibers formation. These results allow to confirm the implication of the new identified genes in the merge of myocytes.
