Comment assurer l’exposition adéquate d’une population d’agneaux lors de l’administration d’antibiotiques via l’eau de boisson en atelier d’engraissement ovin

Abstract : To prevent the development of infectious diseases in sheep fattening houses, antibiotics are systematically administered in food during the fattening start phase. However, preventive treatments should be avoided due to their potential role in the selection of antimicrobial resistance in food-producing animals. One alternative is the metaphylactic use of antibiotics consisting in a collective treatment as soon as the disease is detected in some animals. The collective delivery of antibiotics via drinking water is a promising strategy in this context, but the actually ingested doses are dependent on the individual drinking behaviors which could led to an under exposure of some animals to the antibiotics potentially responsible for therapeutic failures. Thus, we proposed to develop a method to optimize the collective delivery of antibiotics via drinking water. It is presented here for the association of two antibiotics currently used in sheep fattening houses in France, the sulfadimethoxine (SDM) / trimethoprime (TMP) association. As a first step, we determined in our lab the pharmacokinetic parameters of SDM and TMP after an oral administration in lambs just after weaning (pre-ruminating lambs) and two weeks later (ruminating lambs) As a second step, in a sheep fattening house, we administered the SDM/TMP association via drinking water to 200 lambs of about 40 days. We determined the SDM and TMP concentrations in plasma and in waterers and, in the same time, the individual real-time water consumption during the treatment in order to characterize the interindividual variability of plasma concentrations and of water consumption. Ultimately, using the data obtained from the experiments conducted in our laboratory and in the fattening house, a pharmaco-statistical model would be built by incorporating the time series of water consumption in a classical pharmacokinetic model that will be able to predict plasma concentrations actually observed in fattening lambs. Such model will allow to predict for any antibiotic the impact of individual drinking behaviors on individual plasma exposure, and to propose improved dosage regimens adapted to the specificity of animal behaviors, whatever the animal species
