First output from a non-target screening interlaboratory trial on the use of passive samplers for the evaluation of water contamination

Abstract

This study presents the first outputs from an interlaboratory trial, organized within the Norman network (https://www.norman-network.net), where HLB disk passive samplers were exposed for 2 and 4 days in a European river as well as in the output of the local drinking water treatment plant. The disks were desorbed in a laboratory and aliquoted to be sent to the 21 laboratories participating in the trial. In addition, two other samples were prepared: a field blank and a mix of standards in order to build a retention time index. The samples were all analyzed with a column (several possibilities), a gradient and data acquisition parameters imposed by the organizers for a better data interpretation. The first overview given here focuses on four laboratories that acquired data with the same instrument type (QTOF) from a single vendor. Data were processed using the software “MZmine” and the platform “Workflow4metabolomics” but only 9 to 20% of features overlapped between both softwares. For the rest of the study “MZmine” was arbitrarily chosen to perform data processing. While some features were “gained” with a longer exposure time of the passive sampler, other features were lost, revealing an unexpected phenomenon of competition. Overall, the number of features extracted was strongly laboratory dependent. However, all the labs detected much fewer features in drinking water than in river water. The current study reveals that comparing HRMS data (10 to 14% of overlap only between two laboratories) remains extremely complex even though the acquisition was performed on instruments from a single vendor and with a given set of conditions.
