For many years, knowledge on thiol precursors has been limited to S-conjugates of glutathione (G3SH), cysteine (Cys3SH), and later on the dipeptides y-GluCys and CysGly. In this work, we took the parallel between precursor degradation and the glutathione-mediated detoxification pathway a step further by considering a new type of derivative, 3-S-(N-acetyl-Lcysteinyl)hexanol (NAC3SH). This compound was synthesized and then added to the existing liquid chromatography with tandem mass spectrometry (LC-MS/MS) method of thiol precursors. This intermediate was only identified during alcoholic fermentation in synthetic must spiked with G3SH (1 mg/L or 2.45 mu mol/L) in the presence of copper with concentration above 1.25 mg/L, which demonstrates for the first time the existence of this new derivative (until 126 mu g/L or 0.48 mu mol/L) and the capacity of the yeast to produce such a compound. Its status as a precursor was also studied during fermentation, in which a release of 3sulfanylhexanol was noted corresponding to a conversion yield close to 0.6%. This work completed the thiol precursor's degradation pathway in Saccharomyces cerevisiae in synthetic conditions with a new intermediate, confirming its connection with the xenobiotic detoxification pathway and giving new insights on the precursor's fate.
Pendant de nombreuses années, les connaissances sur les précurseurs du thiol se sont limitées aux conjugués S du glutathion (G3SH), de la cystéine (Cys3SH) et, plus tard, des dipeptides y-GluCys et CysGly. Dans ce travail, nous avons poussé le parallèle entre la dégradation des précurseurs et la voie de détoxification médiée par le glutathion un peu plus loin en considérant un nouveau type de dérivé, le 3-S-(N-acétyl-Lcysteinyl)hexanol (NAC3SH). Ce composé a été synthétisé puis ajouté à la méthode existante de chromatographie liquide par spectrométrie de masse en tandem (LC-MS/MS) des précurseurs thiols. Cet intermédiaire n’a été identifié lors de la fermentation alcoolique dans du moût synthétique enrichi de G3SH (1 mg/L ou 2,45 mu mol/L) qu’en présence de cuivre avec une concentration supérieure à 1,25 mg/L, ce qui démontre pour la première fois l’existence de ce nouveau dérivé (jusqu’à 126 mu g/L ou 0,48 mu mol/L) et la capacité de la levure à produire un tel composé. Son statut de précurseur a également été étudié lors de la fermentation, au cours de laquelle une libération de 3sulfanylhexanol a été notée correspondant à un rendement de conversion proche de 0,6%. Ces travaux ont complété la voie de dégradation du précurseur thiol chez Saccharomyces cerevisiae dans des conditions synthétiques avec un nouvel intermédiaire, confirmant son lien avec la voie de détoxification xénobiotique et donnant de nouvelles informations sur le devenir du précurseur.
