Information processing circuits in cell-free systems: beyond gene expression regulation

Cell-free expression systems have emerged as promising tools for implementing information-processing circuits. Most of these circuits employ biosensors directly linked to gene expression regulation, using digital logic encoded on plasmid DNA. Here, we present several new approaches for building biological circuits that process information via metabolic transducers: combining analogue and digital logic in the metabolic perceptron, routing metabolic sensing through a common hub molecule, and screening transducer enzymes using linear DNA templates. The examples introduced here lay the groundwork for more advanced metabolic circuits for rapid and scalable multiplex sensing.
