Endogenous gibberellins in Douglas-fir shoots

Abstract

Endogenous gibberellins (GAs) were identified in shoots from 4-year-old Douglas-fir (Pseudotsuga menziesii) cuttings collected at the floral differentiation time. After purification of shoot extracts on two different HPLC systems (reverse and normal phase), GA-like compounds were detected by radioimmunoassay with polyclonal antibodies raised against GA4 and by the Tanginbozu dwarf rice micro-drop bioassay. GA1, GA3, GA4, GA7 and GA9 from GA-like fractions, and GA9 after enzymatic degradation of a putative GA conjugate -like fraction were identified by combined gas chromatography -mass spectrometry (GC-MS). Full scan mass spectra were obtained for GA4, GA7, GA9 and for the free GA9 of the putative conjugate. The identification of GA1 and GA3 was based on selected ion monitoring and Kovats retention index. GA4, GA7 and GA9 were the predominant gibberellins in Douglas-fir shoots.
