Mécanismes moléculaires de plasticité du système olfactif chez les rongeurs

Abstract : My studies are dedicated to the relationship between olfaction and nutrition in rodents. I’m interested by the plasticity of the olfactory system (mucosa and bulb) under the effect of either short- or long-term modification of nutritional cues, such a fasting or obesity using cellular, molecular and behavioral approaches. After having demonstrated the presence of various hormones involved in the control of food intake (leptin, insulin and their receptors) at several levels of the olfactory system, I’ve analyzed the variation of their expression levels in different nutritional models linked to short-term nutritional plasticity. Transcriptomic analyses have demontrated that fasting modulates the expression of several genes encoding various functions in the olfactory mucosa associated with variations in the odorant responses. Moreover, the olfactory bulb activity is also modulated by nutritional hormones and some olfactory-based behaviors are affected by a short-term manipulation of the endocrine status of animals. My perspectives are to further analyse the consequences of long-term variations of the neurodocrine status on the olfactory capabilities and to decipher the molecular mechanisms responsive of the modulation at the olfactory system periphery : identification of signaling pathways crosstalks, local translation in the apical region of the mucosa and role of the olfactory bulb in the integration of the peripheral information.
