Vers l’obtention de variétés de vigne résistantes à l’oïdium et au mildiou par les méthodes conventionnelles et biotechnologiques

Abstract : The downy and powdery mildews are the two main fungus diseases of grapevine. Though efficiency of copper and sulphur against them has been discovered at the end of the XIXdth century, and considerable progress has been made since in the phytosanitary protection of the vineyards, itscost is high and these two diseases are often uneasy to control. In 1974, INRA undertook to select resistant varieties by the introgression in Vitis vinifera of genes issued from the genetically distant species Muscadinia rotundifolia. One gene, named Run1, conferring total resistance to powdery mildew has been introduced by 6 successive back-crosses of a F1 hybrid. The advanced genotypes bearing the Run1 gene are now under field evaluation for their qualitative and cultural aptitudes. Interestingly, these genotypes showed a partial resistance to downy mildew, however the involvement of the Run1 gene in this phenomenon is still unknown. Work is in progress to introduce in V; vinifera other genes of M. rotundifolia confering hith resistance to downy mildew. In the aim to transfer the powdery mildew resistance to traditional Vitis vinifera varieties, the cloning and sequencing of the Run1 gene have been undertook in collaboration with the australian CSIRO. To ensure its durability against variations of the pathogen, this monogenic resistance will be associated with a partial resistance obtaine by genetic transformation, using genes encoding for PR (pathogenesis-related) proteins capable of disturbing the development of the fungus
