Development and validation of a new HPLC-ESI-MS/MS method, and comparison between on-line and off-line SPE for the analysis of glyphosate and AMPA in freshwaters
Développement et validation d'une méthode HPLC-ESI-MS/MS avec ou sans préconcentration SPE en ligne pour l'analyse du glyphosate et de l'AMPA dans les eaux douces

A new C18 column with Ethylene Bridged Hybrid (BEH) particles was used for the determination of 9-fluorenylmethylchloroformate (FMOC) derivatives of glyphosate and its main metabolite aminomethylphosphonic acid (AMPA) by high performance liquid chromatography coupled to electrospray tandem mass spectrometry (HPLCESI-MS/MS). High efficiency and sensitivity were obtained by using high pH mobile phase buffer that consists of triethylamine (TEA) adjusted with acetic acid. The analytical method involves a sample treatment, a derivatization step and an off-line solid-phase extraction (SPE) sample clean-up/enrichement. An alternative on-line SPE method was developped and the performance of the two approaches were compared.
