Combining ecophysiological and microbial ecological approaches to assess interaction between genotypes of Medicago truncatula and the soil bacterial communities

Résumé

Microbial communities can play a key role in sustainable agriculture by improving crop growth. Currently, a major stake is to study the effect of the plant genotype on the shaping of the soil microbial communities in relation to the plant nutrition. Methods: The links between the plant genotype and the rhizospheric bacterial communities were assessed at both structural and functional level using a multidisciplinary approach. On the one side, the genetic structure (Automated Ribosomial Intergenic Spacer Analysis (ARISA)) and diversity (454 pyrosequencing) of bacterial communities of the rhizosphere was first analysed. On the other side, using an ecophysiological model plant growth was measured dynamically and plant carbon and nitrogen nutrition was estimated at the plant level. Results: The M. truncatula genotype had an effect on the soil bacterial communities at an early developmental stage of plants. At this early stage, while growth of the M. truncatula genotypes was not extensively contrasted, their carbon and nitrogen nutritional strategies differed. Conclusion: These results demonstrate that combining exploration of the soil bacterial communities with an analysis based on an ecophysiological model is a strong tool to characterize the different interactions occurring between plants and soil bacteria. Les communautés microbiennes peuvent jouer un rôle majeur dans le cadre d’une agriculture durable en améliorant la croissance et la santé des plantes. Actuellement, un des enjeux majeur est d’étudier l’effet du génotype de plante sur les communautés microbiennes du sol en relation avec la nutrition de la plante. L’interaction plant – communautés microbiennes de la rhizosphère a ainsi été évaluée par une approche multidisciplinaire. Dans un premier temps, la structure génétique ainsi que la diversité des communautés bactériennes ont été analysées respectivement par DNA fingerprint (ARISA) et pyroséquençage (454). Puis, les différentes stratégies nutritionnelles de la plante ont été analysées par trois stratégies écophysiologiques complémentaire (description phénotypique, analyse des flux, utilisation d’un cadre d’analyse de modèle écophysiologique).A un stade précoce du développement de la plante, le génotype de Medicago truncatula affecte les communautés bactériennes du sol alors que peut de différences phénotypiques ont pu être observées entre les différents génotypes. Cependant, les différents génotypes de plante présentent des stratégies nutritionnelles contrastées.
