Poplar cambium and leaf tissues present specific changes in 2D-page patterns in response to drought of heat stress

Résumé

Stress tolerance of plants is conditioned by their ability to preserve the function of their tissues and cells. The mechanisms involved in the metabolism defence partly depend on the physiological status and on the stress sensing and signalling pathways. Accordingly, stress tolerance arises through molecular responses the patterns of which rely on cell and tissue structures. Thus specific proteomic changes can be expected to occur in leaves and in cambium. Young poplar cuttings grown on soil in 10L pots under controlled conditions were submitted to drought stress by withholding water during 12 days. The progress of the constraint was assessed by measurement of predawn water potential. Plants were then re-watered for a 7 days period. Samples collected on day 8 and 12 corresponded to mild and strong stress, respectively. The heat stress was studied under two contrasting situations. The heat shock treatment consisted in a rapid increase of temperature from 22°C to 42°C, followed by a 48h exposure at 42°C. The second experiment corresponded to a stepwise increase in temperature (25°C during 48h, 30°C during 24h, then 42°C during 48h). In all treatments, a recovery period of 7 days under normal conditions was completed. A sampling was also carried out at this date. TCA-acetone extracted proteins of leaves were separated and quantified by 2D-DIGE. Proteins of interest determined by statistical tests with DeCyder software were identified using a MALDI-Tof-Tof analyser. Xx and XX differential protein spots were stirred up by water or heat stress in leaves and cambial zone respectively. XX% of the altered protein functions were tissue-specific. Beyond stress generic response involving chaperons and antioxidant-linked proteins, results draw up more specific changes. Photosynthesis regulation oriented leaf response and a storage management oriented cambium response are observed.
