Use of fibrolytic enzymes in dairy goats. In vitro evaluation of activity and fermentative characteristics

Utilizacion de enzimas fibroliticas en cabras lecheras. Evaluacion de su actividad y caracteristicas fermentativas in vitro

Résumé

A sequence of in vivo and in vitro experiences were carried out in order to evaluate the effects of supplementing dairy goat diets with fibrolytic enzymes. In the lactation experience (Exp.1) the performance of the main figures of feed intake (DMI), milk production (MP) and milk composition (MC), and the change of body weight (BW) and body condition score (BCS) were studied by a ‘Crossover’ experiment design with 24 Murciano-Granadinas dairy goats in mid lactation, randomly allocated in two treatments (Control: unsupplemented, CON, n = 12; and Promote™: supplemented with fibrolytic enzymes at 0.47 ml/kg DM of concentrate, PRO, n = 12). At the end of the lactation trial (wk 26), eight goats (four per treatment) were selected and allocated in metabolic crates in order to measure the total tract digestibility from wk 27 to 30 of lactation; in vivo digestibility was tried to support with an in vitro assay, determining the effects on gas production profile (GP) and dry matter (DMD) or neutral detergent fiber (NDFD) degradabilities of the same diets offered in vivo (Exp. 2). The GP potential, the cell wall components degradability (NDF and ADF) and the fermentative characteristics of an array of F:C ratios of diets, with different NDF and ADF contents, were also studied (Exp. 3), with two sources of inoculun (dairy cows, DC; and dairy goats, DG) and different doses of PRO enzyme preparation (0, 1 and 2 mg/g of substrate DM) using the cumulative GP technique proposed by Theodorou et al. (1994). The ‘Exp. 4’ was developed with the aim to check the effect of factor variations (pH: 4.0, 5.5, 6.5; temperature: 30, 40, 50 and 70ºC; and enzyme addition dose: 0, 1 and 2 mg/g of substrate DM; and their interactions), on the cellulolytic activities (EA) of two liquid commercial products (PRO and Cellupract AL-100, CEL); the technique of reducing sugars (RS) of Somogyi (1952) was used for it. For data analysis and processing, the GLM and PROC MIXED procedures of SAS (v. 8.1) was used. The DMI, milk yield, 4% fat corrected milk yield, and milk composition were not affected (Exp. 1) by enzyme supplementation, although milk casein content tended to decrease in the enzyme treatment. Body weight change and body condition score change tended to be higher with the enzyme treatment. Digestibilities of DM (CON, 68.9%; PRO, 71.9%) and organic matter (CON, 70.4%; PRO, 72.9%) were higher with enzyme supplementation, while digestibility of N, NDF and acid detergent fiber were not affected with the enzyme addition. Total tract digestibility results could not be supported by the in vitro trial on which similar values were observed for DM and NDF degradability and gas production in both diets. The GP kinetics were affected (P < 0.05) by F:C ratio and was positive determined by the concentrate level in the diet (HF < HMF < LMF < LF) and by source of inoculum (DG > DC; P < 0.001). Enzyme supplementation tended to affect (P < 0.10) or positively affected (P < 0.05) GP profiles determined and predicted (A) in forage diets (HF and HMF) with the average concomitant increase and decrease of constant rates (a and b) and lag time (L), respectively. The in vitro NDFD and ADFD were depressed (P < 0.05) by the increase of concentrate level in the TMR substrates fermented with DC, meanwhile, with DG inoculum, under this conditions, this figures were not affected but even enhanced. The PRO and CEL activity was proved (Exp. 4) to be affected (P < 0.05) in similar way with variation of pH, temperature and addition doses. Maximum EA were found for pH 4.0, 50ºC and the greatest enzyme dose (3 mg/g of substrate DM). Significant interactions were detected among the studied factors. Optimum conditions for product EA expression, found in ‘Exp. 4’, as well as the lack of positive effects on in vivo or in vitro cell wall degradation, DMI and dairy goat productive performance, argue the statements what assure that effectiveness of using fibrolytic enzyme preparations in ruminant nutrition is determined by a whole of related and complex factors, which needs to continue being studied.
En una serie de experiencias in vivo e in vitro, se evaluaron los efectos de la suplementación con enzimas fibrolíticas a la ración de cabras lecheras. En la experiencia de lactación (Exp.1) se estudió, mediante un diseño ‘Crossover’ con 24 cabras Murciano-Granadinas a mediados de lactación y distribuidas en dos tratamientos (Control: sin suplementación, CON, n = 12; y Promote™: suplementadas con enzimas fibrolíticas a 0.47 ml/kg de MS en el concentrado, PRO, n = 12), el comportamiento de los principales indicadores de consumo de alimento (MSI), producción (PL) y composición de la leche (CL), y los cambios en peso vivo (PV) y condición corporal (CC) de los animales. Finalizada la ‘Exp.1’ (semana 26 de lactación), se seleccionaron y alojaron en jaulas de metabolismo 8 cabras representativas (4 por tratamiento) para medir (semana 27 a 30) en un diseño completamente aleatorizado la digestibilidad de las raciones en el tracto total, comprobándose con un ensayo in vitro, los efectos sobre la producción de gas (GP) y la degradabilidad de la materia seca (DMS) y de la fibra neutral detergente (DFND) de las raciones suministradas in vivo (Exp. 2). El potencial de GP, la degradabilidad de los componentes de la pared celular (FND y FAD) y las características fermentativas de un rango de relaciones F:C, con diferentes contenidos en FND y FAD, fueron también estudiados (Exp. 3), con dos fuentes de inoculo (vacas, DC; y cabras lecheras, DG) y diferentes dosis de PRO (0, 1 y 2 mg/g MS sustrato) mediante la técnica propuesta por Theodorou et al. (1994). La ‘Exp. 4’ se desarrolló con el objetivo de comprobar, mediante la técnica de los azúcares reductores (Somogyi, 1952), el efecto de la variación de un rango de factores abióticos (pH: 4.0, 5.5, 6.5; temperatura: 30, 40, 50 y 70ºC; y dosis de adición: 0, 1 y 2 mg/g MS sustrato; y sus interacciones) sobre la actividad celulolítica (AE) de dos productos comerciales (PRO y Cellupract AL-100, CEL). Para el procesamiento y análisis de los datos se utilizaron los procedimientos GLM y PROC MIXED de SAS (v. 8.1). La MSI, la PL (real o corregida a 4% de grasa), y la CL, no fueron afectados (Exp. 1) por la suplementación enzimática, aunque la caseína y el PV y la CC tendieron a disminuir y a aumentar (P < 0.10) con PRO, respectivamente. La suplementación enzimática aumentó (Exp. 2) la DMS (CON, 68.9%; PRO, 71.9%), la digestibilidad de la materia orgánica (DMO) (CON, 70.4%; PRO, 72.9%), y no afectó la DFND, la DFAD y la digestibilidad de la PB; el primer ensayo in vitro, no pudo confirmar las tendencias obtenidas en las condiciones in vivo. La cinética de GP fue afectada (P < 0.05) por la relación F:C y estuvo positivamente determinada por el nivel de concentrado en la ración (HF < HMF < LMF < LF) y por la naturaleza del inoculo (DG > DC; P < 0.001). La suplementación con enzimas tendió a afectar (P < 0.10) o afectó (P < 0.05) positivamente los perfiles de GP real y potencial (A) en las raciones más forrajeras (HF y HMF) con un incremento promedio acompañado de las constantes de ritmo de fermentación (a y b) y la disminución promedio del tiempo lag (L). Las degradaciones in vitro de FND y de FAD fueron disminuidas (P < 0.05) por el aumento de la proporción de concentrado en los sustratos fermentados con DC, mientras que en DG no se afectaron, o incluso se vieron estimuladas en estas condiciones. Se comprobó (Exp. 4) que la AE de PRO y CEL se afecta (P < 0.05) de manera similar cuando las condiciones de pH, temperatura y dosis de aplicación varían. Las máximas AE se obtuvieron para pH 4.0, temperatura 50ºC y la mayor dosis de aplicación (3 mg/g de MS). Se observaron además interacciones significativas entre los factores experimentales estudiados. Las condiciones óptimas obtenidas en la ‘Exp. 4’ para la expresión de la actividad celulolítica de los productos, así como la carencia de efectos positivos sobre la degradación de los componentes de la pared celular in vivo o in vitro, o sobre el consumo y comportamiento productivo de las cabras, argumentan el planteamiento de que la efectividad de estos preparados enzimáticos en la nutrición de rumiantes está predeterminada por un conjunto de factores complejos relacionados entre sí que requieren continuar siendo estudiados.
