Caractérisation de nouveaux microARNs spécifiques de l’ovaire chez le Medaka (Oryzias latipes)

Abstract : The production of good quality gametes is crucial for reproduction. It relies on numerous cellular and molecular intra-ovarian mechanisms during oogenesis. Some of these processes have already been studied in the past. However the role of small non-coding RNA in this function remains still poorly known. In order to determine the sequences and their role in the medaka, a small RNA seq has been done on several specie’s tissues and a 6876 sequences list has been established. The aim of this study is to i) establish a candidate list with an ovary predominant profile, ii) validate the profile of 23 of them, iii) delete the expression of one of the miRNA. All in all 559 candidates were isolated from the Prost ! file and the 23 have been selected on different criteria (profile, annotation and structure prediction). The validation has been done on cDNA of n=11 tissues; 9 somatic but not gonadal (fin, kidney, liver, gill, intestine, brain, eye, muscle, heart) and 2 gonadal (ovary and testis). The profile of 12 genomic locations was analyzed; 5 of them correspond to the sequencing one, 4 have differences and 3 of them are completely different. These data give arguments for the use of the Prost! file in order to determine new candidates. However they will have to be individually validated by qPCR before being chosen for CRISPR inactivation.
